Confocal super-resolution microscopy based on a spatial mode sorter

چکیده
Spatial resolution is one of the most important specifications an imaging system. Recent results in quantum parameter estimation theory reveal that arbitrarily small distance between two incoherent point sources can always be efficiently determined through use a spatial mode sorter. However, extending this procedure to general object consisting many remains challenging, due intrinsic complexity multi-parameter problems. Here, we generalize Richardson-Lucy (RL) deconvolution algorithm address challenge. We simulate its application confocal microscope, with Zernike sorter replacing pinhole used conventional microscope. test different spatially objects arbitrary geometry, and find enhancement sorter-based microscopy on average over 30% higher than microscope using standard RL algorithm. Our method could potentially diverse applications such as fluorescence astronomical imaging.

Confocal reflection microscopy: the "other" confocal mode.
When most biomedical researchers think “confocal microscopy”, they usually have fluorescence imaging in mind. There is a very good reason for this connection. Many biomedical applications of the confocal microscope have utilized its optical sectioning power, combined with the exquisite specificity of immunofluorescence or fluorescence in situ hybridization (FISH) to produce improved images of m...

In traditional optical imaging systems, the spatial resolution is limited by the physics of diffraction. The information on sub-wavelength features is carried by evanescent waves, never reaching the camera, thereby posing a hard limit on resolution: the so-called diffraction limit. Modern microscopic methods enable super-resolution, by employing florescence techniques. Stateof-the-art localizat...

Much analysis of microscope image formation is based upon the so-called weak scaucring approximation in which an image is linearly related to an object by a convenient convolution property which greatly simplifies reconstruction. This approximation is often invalid in tmnsmission microscopy. A better model of transmission must account for multiple scauered components in the light distribution. ...
متن کاملSuper-Resolution Localization Microscopy
Light microscopy techniques have been vital to our understanding of biological structures and systems since their invention in the late 16 Century. However, conventional light microscopy techniques are limited by the diffraction limit of light, preventing us from seeing anything smaller than approximately 200 nm in x and y and approximately 500 nm in z. Many molecules and structures of interest...
